Forskolin, the active compound from Coleus forskohlii root, has been studied for its potential impact on fat metabolism and weight management. Its purported mechanism involves activating the enzyme adenylate cyclase, which increases levels of cyclic AMP (cAMP) in cells. Elevated cAMP is thought to stimulate lipolysis, the breakdown of stored fats.
Some small clinical trials have investigated the effects of forskolin on body composition. For example, a 2005 randomized, double-blind, placebo-controlled study in overweight and obese men found that 250 mg of 10% forskolin extract twice daily for 12 weeks led to a significant reduction in body fat percentage and fat mass compared to placebo, though body weight was not significantly reduced. Other studies in women and other populations have shown mixed or minimal effects, and sample sizes have often been small.
Systematic reviews and meta-analyses suggest that while forskolin may have a mild effect on fat mass, the overall evidence is limited by the low quality and size of available studies. There are also potential side effects, including low blood pressure and interactions with medications.
