8-Prenylnaringenin is a phytoestrogen found predominantly in hops (Humulus lupulus) and is considered one of the most potent plant-derived estrogens identified to date. Scientific studies have demonstrated that 8-prenylnaringenin binds to both estrogen receptor alpha and beta, exhibiting significant estrogenic activity in vitro and in animal models (Milligan et al., 2002). Several small human clinical trials and pilot studies have investigated hop extracts standardized for 8-prenylnaringenin content for the relief of menopausal symptoms, such as hot flashes, which are often associated with low estrogen levels. While some trials have reported modest improvements in symptoms, the evidence base remains limited by small sample sizes and short durations (Erkkola et al., 2010). No large-scale, long-term randomized controlled trials have been conducted to directly demonstrate that 8-prenylnaringenin raises endogenous estrogen levels or fully counteracts the effects of estrogen deficiency. However, the mechanism of estrogen receptor modulation is well established, lending biological plausibility to its use in supporting low estrogen conditions, especially in menopause. More robust clinical data are needed to confirm efficacy and safety, but current evidence provides a moderate scientific rationale for its use in this context.
