Caryophyllene, specifically β-caryophyllene, is a natural bicyclic sesquiterpene found in many essential oils, especially clove oil, and is notable for selectively binding to the cannabinoid receptor type 2 (CB2). Its potential use for epilepsy is based largely on preclinical animal studies rather than longstanding traditional use or robust clinical evidence.

Several studies in rodent models have shown that β-caryophyllene exhibits anticonvulsant effects. For example, research published in Epilepsy & Behavior (2014) demonstrated that β-caryophyllene reduced the severity and duration of seizures in mice induced with pentylenetetrazole (PTZ), a common method for modeling epilepsy. The suggested mechanism involves CB2 receptor activation, which is believed to modulate neuroinflammation and neuronal excitability, both relevant to epileptogenesis.

However, there are currently no published human clinical trials that directly evaluate β-caryophyllene as a treatment for epilepsy, and its efficacy and safety in humans remain unproven. The evidence is therefore limited to mechanistic and animal studies, which justify cautious scientific interest but not clinical endorsement. Thus, while β-caryophyllene is not a traditional epilepsy remedy, there is some emerging scientific rationale for further investigation, primarily from preclinical data.
