Evidence supporting the use of: Papain
For the health condition: Edema
Synopsis
Source of validity: Traditional
Rating (out of 5): 1
Papain, an enzyme derived from the papaya (Carica papaya), has a long history of use in traditional medicine systems, particularly in tropical regions where papaya is native. It has been traditionally used for its proteolytic (protein-digesting) properties to aid digestion, reduce inflammation, and promote wound healing. In the context of edema, traditional medicine practitioners have used papain as part of topical preparations or, less commonly, oral supplements, with the goal of reducing swelling and improving circulation. The rationale is that its enzymatic activity could help break down inflammatory proteins and facilitate fluid resorption from tissues.
However, scientific evidence supporting papain’s effectiveness in treating or supporting edema is limited. Some proteolytic enzymes, like bromelain, have shown mild anti-inflammatory effects in preliminary studies, but papain itself has not been robustly studied for this specific indication. Clinical trials are scarce, and those that exist often include papain in combination with other enzymes, making it difficult to attribute effects solely to papain. The bulk of evidence for papain’s use in edema thus stems from traditional use rather than rigorous scientific validation. Its oral use is further limited by concerns about allergenicity and potential gastrointestinal irritation. Overall, while papain is traditionally used for edema, the quality and quantity of scientific evidence supporting this practice is weak.
