Vitamin B6 (pyridoxamine HCl), as well as other forms of vitamin B6, has some scientific basis for use in supporting or treating depression, although the evidence is limited and not robust. Vitamin B6 is involved in the synthesis of neurotransmitters such as serotonin, dopamine, and gamma-aminobutyric acid (GABA), all of which play a role in mood regulation. Several observational studies have found that individuals with low levels of vitamin B6 are more likely to experience depressive symptoms. Some small-scale clinical trials have explored vitamin B6 supplementation as an adjunct to standard antidepressant therapy or in populations with B6 deficiency showing modest improvements in mood. However, results are mixed, and large, well-designed randomized controlled trials are lacking.
A 2022 double-blind placebo-controlled study (Parry et al., 2022) found that high-dose vitamin B6 supplementation reduced self-reported anxiety but not depression scores in young adults. Earlier meta-analyses and reviews suggest a potential benefit for B-vitamin supplementation (including B6) in individuals with low baseline levels or elevated homocysteine, but little effect in the general population. Overall, while there is a plausible biochemical rationale and some clinical support, the strength of evidence is moderate to low, justifying a rating of 2. Vitamin B6 supplementation may be most useful in those with a demonstrated deficiency rather than as a general treatment for depression.
