Prebiotics, defined as non-digestible food components that beneficially affect the host by selectively stimulating the growth or activity of certain gut bacteria, have garnered attention for their potential to influence mental health, including depression. The gut-brain axis—a bidirectional communication system between the gastrointestinal tract and the central nervous system—provides the biological rationale for exploring prebiotics in this context. Some studies, including randomized controlled trials, have shown that certain prebiotics (e.g., galacto-oligosaccharides or fructo-oligosaccharides) can modulate the composition of gut microbiota, which in turn may impact mood and depressive symptoms through immune, neural, and endocrine pathways. However, while there is emerging evidence from animal studies and small human trials suggesting that prebiotics may reduce markers of stress and improve mood, the overall quality and quantity of evidence remain limited. Most human research is preliminary, with small sample sizes, short durations, and heterogeneity in both the type and dose of prebiotics used. Meta-analyses and systematic reviews note that while there is promise, more robust and well-powered studies are needed to confirm efficacy and clarify mechanisms. Therefore, while scientific interest and some initial data exist, the evidence base supporting the use of unspecified prebiotics for depression is currently weak to modest.
