Phenylethylamine (PEA) is a naturally occurring trace amine found in the human body and in certain foods. It acts as a central nervous system stimulant, with a structure similar to amphetamine. There is some scientific interest in PEA for its potential to alleviate symptoms of depression, as it may increase the availability of neurotransmitters such as dopamine and norepinephrine, which are often implicated in mood regulation. Some small clinical studies and case reports have suggested that oral or supplemental PEA, especially when combined with monoamine oxidase B (MAO-B) inhibitors (which prevent its rapid breakdown), can produce antidepressant effects in patients with refractory depression.

However, the quality and quantity of the available scientific evidence are limited. Most clinical data come from small, open-label studies rather than large-scale, randomized controlled trials. In these studies, PEA supplementation (often combined with selegiline, an MAO-B inhibitor) appeared to provide rapid-onset antidepressant effects in some patients. Nonetheless, the effect size, safety, and long-term efficacy remain uncertain. The rapid metabolism of PEA in the body also limits its effectiveness unless combined with MAO-B inhibition. Overall, while there is some preliminary scientific support for using PEA in depression, the evidence base is weak and further research is needed.
