Ligustilide is a major bioactive component found in several traditional Chinese medicinal herbs, most notably Angelica sinensis (Dong Quai). Historically, these herbs have been used in traditional Chinese medicine (TCM) to "nourish the blood," regulate menstruation, and support emotional well-being, which has sometimes included symptoms overlapping with what is now considered depression. However, the direct traditional use of ligustilide itself for depression is not well documented; rather, the whole herb or herbal formula is used, with ligustilide as one of its constituents.

Modern scientific studies investigating ligustilide’s effects on depression are limited, mostly preclinical and in animal models. Some animal studies suggest that ligustilide may have neuroprotective, anti-inflammatory, and anti-oxidative properties, and a few rodent studies indicate potential antidepressant-like effects—possibly via modulation of neurotransmitters or reduction of neuroinflammation.
