Ligustilide is a phthalide compound primarily found in herbs like Angelica sinensis (Dong Quai) and Ligusticum chuanxiong, both of which are commonly used in traditional Chinese medicine (TCM). The use of these herbs in TCM for mood support and emotional well-being is well documented, but the tradition is largely based on empirical observation and holistic theories rather than evidence-based research. While ligustilide is one of the main active components of these herbs, there is very limited direct scientific evidence linking ligustilide itself to antidepressant effects in humans. A few preclinical studies in animal models have suggested neuroprotective, anti-inflammatory, and possible antidepressant-like effects of ligustilide, but these findings are preliminary and not sufficient to establish clinical efficacy. No robust human clinical trials exist to support the use of ligustilide specifically for depression. Therefore, while its use is justified by tradition (through the use of the whole herb in TCM), the evidence supporting ligustilide as a treatment or support for depression is weak and primarily historical rather than scientific.
