Alpha glyceryl, more accurately known as alpha-glycerylphosphorylcholine (Alpha-GPC), is a cholinergic compound used as a dietary supplement. It is thought to support cognitive function by serving as a precursor to acetylcholine, a neurotransmitter essential for memory and learning. Several small clinical studies have investigated Alpha-GPC in the context of dementia, particularly Alzheimer's disease and vascular dementia. Some randomized controlled trials, primarily conducted in Italy and published in the 1990s and early 2000s, reported modest improvements in cognitive scores (such as the Mini-Mental State Examination) among dementia patients taking Alpha-GPC compared to placebo. Meta-analyses and reviews have concluded that Alpha-GPC may offer mild cognitive benefits in dementia, but the overall quality of evidence is low to moderate due to small sample sizes, short durations, and methodological limitations.

Alpha-GPC is not as extensively studied as standard treatments like acetylcholinesterase inhibitors, and major clinical guidelines do not recommend it as a primary intervention for dementia. However, its use is scientifically motivated by its mechanism of action and the limited but positive findings from clinical trials. Additional, larger, and more rigorous studies are needed to confirm its efficacy and safety for dementia patients.
