Nicotinamide riboside (NR) is a form of vitamin B3 and a precursor to nicotinamide adenine dinucleotide (NAD+), a molecule essential for cellular energy metabolism and repair. In recent years, scientific interest has grown regarding its potential role in inflammatory bowel diseases such as colitis. Preclinical studies, particularly in animal models, have shown that NR supplementation can ameliorate colitis symptoms. For example, research in mice with chemically induced colitis has demonstrated that NR administration helps restore NAD+ levels, reduce oxidative stress, and limit inflammatory signaling pathways (notably NF-κB). These effects collectively lead to reduced mucosal damage and improved intestinal barrier function. However, evidence in humans is extremely limited. There are no large-scale clinical trials directly investigating NR supplementation for colitis in people. The mechanistic rationale is compelling, as NAD+ depletion is observed in inflamed intestinal tissues and boosting NAD+ with NR may support cellular repair and immune regulation. Nevertheless, without robust human data, the evidence supporting NR's use for colitis remains preliminary. In summary, while there is emerging scientific validation from animal studies, the lack of human clinical trial data makes the current evidence modest (rated 2/5). NR cannot yet be considered a standard or proven therapy for colitis, but ongoing research may further elucidate its potential benefits.
