Tributyrin is a triglyceride form of butyric acid, a short-chain fatty acid (SCFA) that is naturally produced in the colon from the fermentation of dietary fibers by gut microbiota. Butyric acid is a key energy source for colonocytes and has well-documented anti-inflammatory and mucosal healing properties. Scientific interest in tributyrin as a treatment for colitis stems from its ability to deliver butyrate effectively to the colon, overcoming the rapid absorption and metabolism issues associated with free butyric acid.
Several animal studies have demonstrated that tributyrin supplementation can reduce inflammation and promote mucosal repair in models of colitis (such as DSS-induced colitis in mice). The proposed mechanisms include inhibition of nuclear factor kappa B (NF-κB) activation, promotion of regulatory T cell differentiation, enhancement of mucosal barrier integrity, and reduction of pro-inflammatory cytokine production. A few small pilot studies and case reports in humans have suggested possible benefits in inflammatory bowel disease (IBD), but robust randomized controlled trials are lacking.
The evidence base is primarily preclinical, with limited but promising human data. Therefore, while tributyrin shows scientific rationale and some validation for supporting colitis management, the overall evidence is moderate (rated 3 out of 5) and further clinical research is needed to confirm its efficacy and optimal dosing in human patients with colitis.
