Vitamin D (ergocalciferol, also known as Vitamin D2) is scientifically validated for use in supporting bone healing and overall bone health. Vitamin D is essential for the regulation of calcium and phosphate metabolism, both of which are critical for bone mineralization and repair. Several clinical studies and reviews indicate that adequate Vitamin D status is associated with improved fracture healing and reduced risk of complications such as delayed union or nonunion. Patients with Vitamin D deficiency are at increased risk for impaired bone healing and secondary complications such as osteomalacia.

While most research focuses on cholecalciferol (Vitamin D3), ergocalciferol (Vitamin D2) is also effective in raising serum 25(OH)D levels and is used in supplementation, particularly in vegetarians or in individuals with specific dietary preferences. Clinical guidelines from organizations such as the American Academy of Orthopaedic Surgeons recommend assessing and correcting Vitamin D deficiency in patients with fractures, especially in older adults and those at risk for osteoporosis.

However, while supplementation is beneficial in those with deficiency or at risk, there is less evidence that high-dose supplementation improves fracture healing in individuals with adequate baseline Vitamin D levels. Nonetheless, the overall body of evidence supports the use of Vitamin D to optimize bone metabolism and healing processes after fracture, particularly in deficient populations.
