MK-677 (also known as Ibutamoren) is a non-peptide agonist of the ghrelin receptor that stimulates the secretion of growth hormone (GH) and insulin-like growth factor 1 (IGF-1). Both GH and IGF-1 are known to play significant roles in bone metabolism and repair. Preclinical animal studies and limited human trials suggest that MK-677 can increase circulating levels of these hormones, which are associated with enhanced bone turnover and potentially improved fracture healing. However, direct clinical evidence demonstrating that MK-677 improves outcomes in patients with broken bones (fractures) is sparse. Most of the evidence comes from studies on its general anabolic effects, increased bone mineral density in elderly populations, or its potential to counteract muscle wasting. There are no large-scale, randomized controlled trials specifically assessing fracture healing or bone repair in humans treated with MK-677. Therefore, while there is a plausible biological mechanism and some indirect scientific support, the evidence is preliminary and does not yet robustly justify its clinical use for treating broken bones.
