Ibutamoren (MK-677) is a non-peptide growth hormone secretagogue that stimulates the secretion of growth hormone (GH) and insulin-like growth factor 1 (IGF-1). Scientific interest in ibutamoren for bone healing arises from the established role of GH and IGF-1 in bone metabolism and regeneration. Several preclinical studies in animals have demonstrated that ibutamoren can increase bone mineral density and may accelerate fracture healing, likely via enhanced osteoblast activity and improved bone remodeling. In humans, some clinical trials and case reports suggest that long-term administration of ibutamoren increases bone mineral density, particularly in elderly populations or those with growth hormone deficiencies.

However, direct high-quality clinical evidence supporting the use of ibutamoren specifically for treating or accelerating the healing of broken bones (fractures) in humans is limited. Most human studies focus on bone density rather than fracture repair, and existing studies often have small sample sizes or short follow-up periods. As such, while the biological rationale is strong and animal data are promising, the clinical evidence for fracture healing is still preliminary. Therefore, the use of ibutamoren for broken bones is best described as having a scientific basis with modest supporting evidence (rating: 2 out of 5), and more robust clinical trials are needed to confirm its efficacy for this specific indication.
