Coleus forskohlii (and its active compound, forskolin) has gained popularity in the bodybuilding community primarily due to its purported effects on body composition and fat loss. Forskolin is known to activate the enzyme adenylate cyclase, which increases intracellular levels of cyclic AMP (cAMP). Elevated cAMP is associated with increased fat breakdown (lipolysis) and may influence muscle mass via hormonal pathways. Several small-scale human studies have explored forskolin’s effects on body composition. For example, a 2005 randomized controlled trial in overweight and obese men found that 250 mg of Coleus forskohlii extract (10% forskolin), taken twice daily for 12 weeks, resulted in a significant reduction in body fat percentage and fat mass compared to placebo, with a trend toward increased lean body mass. However, subsequent research has produced mixed results: some studies report no significant effects on fat loss or muscle gain, especially in women or in normal-weight individuals.

Overall, the evidence supporting Coleus forskohlii for bodybuilding purposes is limited and inconsistent, and most studies are small, short-term, and of variable quality. There is some scientific rationale and preliminary human data, but it does not strongly support widespread use for muscle growth or fat loss.
