Cardarine (also known as GW501516) is a peroxisome proliferator-activated receptor delta (PPARδ) agonist originally developed for metabolic and cardiovascular diseases. In the bodybuilding community, Cardarine is used primarily for its purported ability to enhance endurance, promote fat loss, and improve lipid profiles. There is some preclinical scientific evidence—mainly from animal studies and limited human trials—that GW501516 can increase fatty acid oxidation, elevate exercise endurance, and improve certain aspects of metabolism. However, robust, peer-reviewed human studies focusing specifically on athletes or bodybuilders are lacking.

Notably, Cardarine was discontinued in clinical development due to concerns about cancer risk in animal studies. Furthermore, its use is banned by the World Anti-Doping Agency (WADA) because of its performance-enhancing effects and potential health risks. To date, evidence supporting Cardarine’s use in bodybuilding is limited and comes primarily from mechanistic studies and anecdotal reports rather than large, controlled human clinical trials. Therefore, while there is a scientific rationale for its purported effects, the evidence is not strong or conclusive, and significant safety concerns remain.
