2a, 17a-dimethyl-17b-hydroxy-5a-androstan-3-one, also known as methyl-1-testosterone or M1T, is an orally active anabolic-androgenic steroid (AAS). Its use in bodybuilding is primarily supported by scientific understanding of its pharmacological effects, not tradition. M1T is a synthetic derivative of dihydrotestosterone (DHT) that exhibits potent anabolic activity. Peer-reviewed studies and anecdotal reports indicate that it significantly increases muscle mass and strength when used in conjunction with resistance training, due to its ability to enhance protein synthesis and nitrogen retention in muscle tissue. As a methylated AAS, it is orally bioavailable, making it attractive for non-injectable use among athletes and bodybuilders.

However, the evidence base is largely extrapolated from general AAS research and limited clinical studies on M1T itself. Its anabolic potency is reported to be several times greater than testosterone, with a relatively low androgenic effect. Despite these effects, M1T use is associated with significant health risks, including hepatotoxicity, suppression of natural testosterone production, negative lipid profile changes, and other serious side effects. M1T was never approved for medical use and has been classified as a controlled substance in many countries. Its use in bodybuilding is banned by most sports organizations. Nonetheless, the scientific rationale for its muscle-building properties is well established, though overshadowed by safety concerns.
