Vitamin D has been studied for its potential role in the prevention of certain birth defects, especially those related to bone development and fetal growth. Scientific interest primarily focuses on the association between maternal vitamin D deficiency and increased risk of skeletal disorders (such as rickets) and possibly other congenital abnormalities. There is also some evidence that vitamin D may influence immune and metabolic processes in early fetal development. However, the strength of the evidence supporting vitamin D supplementation specifically for the prevention of birth defects is limited. Observational studies have shown that low maternal vitamin D levels are associated with increased risk of certain pregnancy complications, like preeclampsia, low birth weight, and impaired skeletal development in the newborn. Randomized controlled trials directly assessing vitamin D supplementation for prevention of birth defects are few, and results are mixed. Professional organizations recommend ensuring adequate vitamin D levels during pregnancy, but this is generally aimed at supporting maternal health and optimal fetal bone development, rather than specifically targeting classic birth defects such as neural tube defects (which are primarily prevented by folic acid). While maintaining sufficient vitamin D status is considered important for overall pregnancy outcomes, routine high-dose supplementation solely for birth defect prevention is not universally recommended. Thus, there is scientific rationale for its use, but robust clinical evidence for birth defect prevention is still emerging.
