L-citrulline is an amino acid commonly used as a dietary supplement to support vascular and arterial health. Its primary mechanism of action involves its role as a precursor to L-arginine, which is subsequently converted into nitric oxide (NO) in the body. Nitric oxide is a key vasodilator, meaning it helps relax and widen blood vessels, thereby supporting healthy blood flow and arterial function.

Multiple clinical studies have demonstrated that L-citrulline supplementation can increase plasma L-arginine levels and enhance nitric oxide production more effectively than L-arginine supplementation itself. Randomized controlled trials suggest that L-citrulline can improve endothelial function, lower blood pressure in prehypertensive and hypertensive individuals, and reduce arterial stiffness. For example, a 2010 study in the American Journal of Hypertension found that L-citrulline supplementation significantly reduced aortic blood pressure and improved arterial stiffness in middle-aged adults. Other studies have also reported improvements in flow-mediated dilation, a marker of endothelial health.

Although the body of evidence is not as extensive as for some pharmaceutical interventions, the mechanistic plausibility and consistent findings from human trials provide scientific validation for the use of L-citrulline in supporting arterial health. However, more large-scale, long-term studies are warranted to fully establish its clinical relevance and optimal dosing.
