7,8-Dihydroxyflavone (7,8-DHF) is a small molecule that acts as a selective agonist of the TrkB receptor, which is the main receptor for brain-derived neurotrophic factor (BDNF). The interest in 7,8-DHF for anxiety disorders stems from robust preclinical research, particularly in animal models, showing that 7,8-DHF can produce anxiolytic-like effects. These effects are likely related to its capacity to enhance BDNF signaling, which plays a key role in synaptic plasticity, neurogenesis, and resilience to stress—all processes implicated in the pathophysiology of anxiety. In rodent studies, administration of 7,8-DHF reduces anxiety-like behaviors in paradigms such as the elevated plus maze and open field test. Additionally, 7,8-DHF has shown potential to reverse anxiety-like symptoms induced by chronic stress and to rescue synaptic deficits in animal models. However, it is important to note that there is currently no published clinical trial data evaluating the efficacy or safety of 7,8-DHF in humans with anxiety disorders. All current evidence is limited to basic science and animal studies. While the mechanistic rationale and preclinical findings are promising, direct scientific validation in human subjects is lacking at this time. Thus, the rating of evidence is moderate but not high, reflecting the gap between animal data and clinical application.
