Vitamin E, particularly in its alpha-tocopherol form, has been investigated for its potential role in the treatment and support of Alzheimer's Disease (AD) due to its antioxidant properties. Oxidative stress is a notable feature in the development and progression of AD, and antioxidants like Vitamin E have been proposed as possible interventions. Several clinical trials have explored this hypothesis. For example, a landmark study published in The New England Journal of Medicine (Sano et al., 1997) demonstrated that high-dose alpha-tocopherol (2000 IU/day) modestly delayed clinical progression in patients with moderate AD compared to placebo. More recently, the TEAM-AD VA Cooperative Randomized Trial (Dysken et al., 2014) found that Vitamin E supplementation was associated with a slower rate of functional decline in patients with mild to moderate AD. However, most of these studies used alpha-tocopherol, not gamma-tocopherol. Gamma-tocopherol is another common form of Vitamin E in the diet, but its effects in AD are less well studied. Moreover, meta-analyses and systematic reviews indicate that the benefits of Vitamin E are modest and must be weighed against potential risks, such as increased mortality at high doses. Current clinical guidelines do not universally recommend Vitamin E for AD, but some clinicians may consider it as an adjunctive therapy. In summary, there is scientific investigation and limited validation for Vitamin E (primarily alpha-tocopherol) in AD, but evidence is moderate and not conclusive, especially for gamma-tocopherol.
