Pomegranate (Punica granatum) is being investigated for its potential role in supporting or treating Alzheimer's Disease (AD), primarily based on emerging scientific evidence rather than traditional use. The interest stems from the fruit’s high content of polyphenols, especially punicalagin and ellagic acid, which exhibit antioxidant and anti-inflammatory properties. Oxidative stress and neuroinflammation are key contributors to the pathogenesis of AD, making pomegranate a candidate for intervention.

Preclinical studies, including in vitro and animal models, have demonstrated that pomegranate extracts and juice can reduce amyloid-beta accumulation, decrease oxidative damage, and improve cognitive function. For example, a 2006 study published in Neurobiology of Disease found that pomegranate juice supplementation reduced amyloid deposition and improved behavioral performance in a mouse model of Alzheimer's. Additionally, some studies suggest pomegranate polyphenols may inhibit the formation of neurotoxic aggregates and modulate signaling pathways involved in neuronal survival.

However, clinical evidence in humans is limited. A few small pilot trials have shown cognitive benefits in older adults or those with mild memory complaints, but these studies are generally underpowered and lack replication. There are no large-scale, definitive clinical trials proving efficacy in diagnosed AD patients. Thus, while the scientific rationale and early data are promising, evidence remains preliminary, and more research is needed before pomegranate can be recommended as an effective therapy for Alzheimer’s Disease.
