Polyphenols are a diverse group of naturally occurring compounds found in plants, including flavonoids, phenolic acids, and stilbenes. Their use to support or treat Alzheimer's Disease (AD) is primarily justified by emerging scientific research rather than traditional medicine. Preclinical studies (in vitro and animal models) have shown that certain polyphenols—such as resveratrol (from grapes), curcumin (from turmeric), and epigallocatechin gallate (EGCG, from green tea)—may have neuroprotective effects. These effects appear to be mediated by their antioxidant, anti-inflammatory, and anti-amyloid properties. For example, polyphenols can scavenge free radicals, reduce neuroinflammation, inhibit amyloid-beta aggregation, and modulate signaling pathways involved in neuronal survival. However, the evidence from human clinical trials is limited and inconsistent. Some small-scale clinical studies have reported modest cognitive benefits or biomarker improvements with polyphenol supplementation, but most have failed to demonstrate significant or reproducible effects in preventing or treating Alzheimer's Disease. Issues such as poor bioavailability, differences in formulations, and small sample sizes complicate interpretation. As a result, while there is a scientific rationale and some preliminary evidence, the overall quality and consistency of the data supporting polyphenols for Alzheimer's Disease is low, reflecting a rating of 2 out of 5. Substantial further research, particularly large-scale, well-designed clinical trials, is needed to determine their true efficacy in humans.
