Polyphenols, a diverse group of naturally occurring compounds found in plants (such as flavonoids, phenolic acids, stilbenes, and lignans), have been studied for their potential neuroprotective effects, including roles in the prevention and treatment of Alzheimer's Disease (AD). The scientific justification for their use is based primarily on preclinical studies and some early-stage clinical research.
Polyphenols are thought to exert beneficial effects through several mechanisms relevant to AD pathology: they possess strong antioxidant properties, can modulate neuroinflammation, inhibit amyloid-beta aggregation (a hallmark of AD), and support neuronal signaling pathways. Compounds such as resveratrol (found in grapes), epigallocatechin gallate (EGCG, from green tea), and curcumin (from turmeric) have demonstrated the ability to reduce amyloid plaque formation and tau pathology in animal and cellular models.
However, while these findings are promising, clinical evidence in humans remains limited and of low quality. Some small randomized controlled trials suggest cognitive benefits or biomarker improvements, but results are inconsistent, and larger, more rigorous studies are needed. Issues such as poor bioavailability of polyphenols and variability in individual responses present further challenges.
