Niacinamide (vitamin B3) has attracted attention for its potential neuroprotective effects in Alzheimer's disease (AD), but scientific validation remains preliminary. The interest stems from research indicating that niacinamide may influence pathways relevant to AD, such as neuroinflammation, mitochondrial function, and tau pathology. Several animal studies have demonstrated that high-dose niacinamide can reduce tau phosphorylation, restore synaptic plasticity, and improve cognitive performance in transgenic mouse models of AD. For example, a widely cited 2008 study showed that niacinamide supplementation reversed cognitive deficits and reduced tau pathology in mice engineered to develop Alzheimer-like symptoms.
However, robust human evidence is lacking. There are currently no large-scale, high-quality clinical trials demonstrating that niacinamide supplementation improves cognition or slows progression in AD patients. Some small pilot studies and case reports have suggested potential cognitive benefits, but these findings are inconclusive and not universally replicated. Furthermore, most human data pertains to general cognitive support in the elderly, rather than AD specifically.
In summary, while there is some scientific rationale and promising preclinical evidence for niacinamide in AD, clinical validation is insufficient at this time. Research is ongoing, and niacinamide is not yet an established or widely recommended therapy for Alzheimer's disease.
