Mangiferin, a polyphenolic compound found primarily in mangoes (Mangifera indica), has been studied for its potential neuroprotective effects relevant to Alzheimer's Disease (AD). Preclinical evidence from cell and animal models suggests that mangiferin exerts antioxidant, anti-inflammatory, and anti-amyloidogenic properties. For instance, studies in rodents have shown that mangiferin can reduce oxidative stress, inhibit acetylcholinesterase (thereby increasing acetylcholine levels), and modulate neuroinflammation—mechanisms implicated in AD pathology. Some research also indicates that mangiferin may decrease beta-amyloid accumulation and tau phosphorylation, both central to AD progression (source). However, despite promising results in laboratory settings, there is a lack of robust clinical (human) trials directly evaluating mangiferin’s efficacy in preventing or treating Alzheimer's Disease. Most available data are limited to in vitro (cell culture) and in vivo (animal) studies, which may not fully translate to human disease. Thus, while the scientific rationale exists and preliminary findings are encouraging, the current level of evidence is low and does not yet support clinical use. More well-designed clinical trials are necessary to establish mangiferin’s safety and effectiveness in humans with Alzheimer's Disease.
