Hericenones are a class of compounds found in the fruiting body of the mushroom Hericium erinaceus (Lion’s Mane). Their use for supporting or treating Alzheimer’s Disease is justified primarily by preliminary scientific evidence, not tradition. Several in vitro and animal studies suggest that hericenones, along with related compounds called erinacines, can stimulate the synthesis of nerve growth factor (NGF), a protein vital for the growth, maintenance, and survival of neurons. Since NGF levels are reduced in Alzheimer’s Disease, boosting NGF has been proposed as a potential therapeutic strategy. Some rodent studies have shown that Hericium erinaceus extracts can improve memory and cognitive function and reduce amyloid-beta plaque accumulation, a hallmark of Alzheimer’s pathology. However, these effects have not been robustly confirmed in large-scale, high-quality human clinical trials. A few small, short-term clinical studies in elderly individuals with mild cognitive impairment have shown modest cognitive benefits, but patients with established Alzheimer’s Disease have not been thoroughly studied. Thus, while there is some scientific rationale and early evidence supporting the use of hericenones for cognitive support, the evidence remains limited and preliminary, and more research—especially in humans—is needed to confirm effectiveness and safety in Alzheimer’s Disease.
