Epigallocatechin gallate (EGCG) is a major polyphenol found in green tea, and its potential neuroprotective effects have attracted scientific interest for Alzheimer's Disease (AD). Preclinical studies in cell and animal models suggest that EGCG may exert beneficial effects via several mechanisms relevant to AD pathology, including inhibition of amyloid-beta (Aβ) aggregation, reduction of oxidative stress, anti-inflammatory action, and modulation of signaling pathways involved in neuronal survival and synaptic plasticity. For example, EGCG has been shown to remodel toxic Aβ oligomers and fibrils into less toxic species, and to reduce Aβ-induced neurotoxicity in vitro. Animal studies have reported improvements in cognitive function and reductions in amyloid plaque burden following EGCG administration.
However, the translation of these findings to human clinical efficacy remains limited. Clinical trials evaluating EGCG in Alzheimer's patients are sparse and have generally been small or focused on surrogate biomarkers rather than cognitive outcomes. A few pilot studies have examined the effects of green tea extracts (containing EGCG) on cognition in elderly populations, but results are inconclusive or show only modest benefits. Safety and bioavailability concerns also limit the clinical application, as EGCG’s absorption in humans is low and high doses may have adverse effects.
In summary, while there is a scientific rationale and promising preclinical data supporting the use of EGCG for Alzheimer’s Disease, robust clinical evidence in humans is lacking. Thus, the current level of scientific evidence is low to moderate, justifying a rating of 2 out of 5.
