Curcuminoids, the active compounds in turmeric (Curcuma longa), have been investigated for their potential role in supporting or treating Alzheimer's Disease (AD). The interest primarily arises from their anti-inflammatory, antioxidant, and anti-amyloid properties, which are relevant because AD is characterized by neuroinflammation, oxidative stress, and accumulation of beta-amyloid plaques in the brain. Preclinical studies, particularly in animal models, have shown that curcuminoids can reduce amyloid plaque burden, inhibit tau phosphorylation, and modulate neuroinflammatory pathways.

However, clinical evidence in humans is limited and inconsistent. Several small-scale clinical trials have explored curcumin supplementation in patients with Alzheimer's or mild cognitive impairment. Most of these studies report minimal to no cognitive benefits, largely due to curcumin’s poor bioavailability. Some formulations with enhanced absorption have shown modest improvements in cognitive function and reductions in amyloid and tau accumulation in imaging studies, but these results are preliminary and not yet widely replicated.

Overall, while there is scientific rationale and promising preclinical data supporting the use of curcuminoids for AD, robust clinical evidence is lacking. Larger, well-designed human studies are needed to determine efficacy and safety. Thus, the scientific validation is weak (rating 2/5), and curcuminoids are not considered an established treatment for Alzheimer's Disease.
