AMP-activated protein kinase (AMPK) is not an ingredient but a cellular enzyme that plays a crucial role in maintaining energy homeostasis. In the context of Alzheimer's Disease (AD), scientific interest in AMPK arises from preclinical studies rather than long-standing traditional use or direct clinical validation. AMPK activation has been shown in cell and animal models to promote neuronal survival, reduce neuroinflammation, and regulate autophagy—processes that are disrupted in AD. Some studies suggest that AMPK activation can decrease the accumulation of amyloid-beta (Aβ) plaques and tau tangles, hallmark features of AD pathology. Furthermore, several pharmaceuticals and natural compounds (like metformin and resveratrol) are known to activate AMPK and have been investigated for their neuroprotective effects.
However, the evidence supporting AMPK activation as a strategy for treating or preventing Alzheimer's Disease in humans remains limited. Most findings are based on laboratory or animal research, with only a few small-scale clinical studies exploring AMPK-activating agents. The direct translation of these findings into clinical benefit for AD patients is not yet established. There is also some controversy, as excessive or dysregulated AMPK activation in certain contexts might contribute to neurodegeneration. In summary, there is scientific rationale and early preclinical evidence for targeting AMPK in AD, but robust clinical validation is lacking, justifying a moderate-low evidence rating.
