Clostridium butyricum is a probiotic bacterium that has been studied for its potential immunomodulatory effects, including its role in supporting or treating respiratory allergies such as allergic rhinitis and asthma. The rationale behind its use is based on its ability to produce short-chain fatty acids (SCFAs), particularly butyrate, which are known to influence immune responses and help maintain intestinal barrier function. SCFAs may promote regulatory T cells (Tregs), which can dampen allergic inflammation. Some preclinical studies in animal models have demonstrated that C. butyricum administration can reduce airway hyperresponsiveness and eosinophilic inflammation, suggesting a potential benefit in allergic airway diseases. A limited number of small clinical studies and case reports in humans suggest that supplementation may alleviate symptoms or modulate immune markers relevant to allergies, but these findings are preliminary and often lack robust controls or large sample sizes.

However, the overall quality and quantity of clinical evidence remain modest. There are no large-scale randomized controlled trials specifically establishing C. butyricum as an effective treatment for respiratory allergies in humans. Most of the existing support comes from mechanistic and animal studies, with only early-stage human research available. Therefore, while there is a scientific rationale and some evidence, it is not yet strong or definitive.
