Vitamin E, particularly in the form of alpha-tocopheryl polyethylene glycol succinate (TPGS), is a derivative designed to enhance the solubility and bioavailability of vitamin E. The main justification for the use of vitamin E in aging prevention lies in its established role as a potent antioxidant. Oxidative stress and free radical damage are widely recognized contributors to cellular aging and age-related diseases. By neutralizing free radicals, vitamin E has been hypothesized to slow the aging process of cells and tissues. Numerous in vitro and animal studies show vitamin E can reduce oxidative damage and modulate cell signaling pathways associated with aging. Observational studies in humans have linked higher dietary intake of vitamin E with reduced risk of some age-related conditions, such as cognitive decline and cardiovascular disease. However, large randomized controlled trials have generally failed to show significant benefits of vitamin E supplementation in preventing age-related diseases or extending lifespan in healthy adults. Some studies have even raised concerns about potential harm at high doses. Regarding TPGS specifically, its use is more common as a drug delivery enhancer rather than as a direct anti-aging supplement. Direct clinical evidence supporting TPGS-vitamin E for aging prevention in humans is lacking. In summary, while the antioxidant rationale for vitamin E in aging is scientifically credible, robust clinical validation for its effectiveness in aging prevention, especially for TPGS forms, is limited.
