Nicotinamide riboside (NR) is a form of vitamin B3 that has gained attention for its potential role in supporting healthy aging and potentially slowing age-related decline. NR is a precursor to nicotinamide adenine dinucleotide (NAD+), a coenzyme that is critical for cellular metabolism, DNA repair, and mitochondrial function—all processes that become less efficient with age. Scientific interest in NR is based on data from animal studies and early-phase human trials showing that supplementation can increase NAD+ levels in blood and tissues. In animal models, boosting NAD+ through NR administration has been linked to improved mitochondrial function, enhanced physical activity, better glucose metabolism, and increased resistance to metabolic stress.
However, evidence supporting the use of NR for aging prevention in humans is still emerging. Some small human studies have demonstrated that NR supplementation can safely elevate NAD+ levels, but clear clinical benefits such as improved physical or cognitive function, longevity, or reduced incidence of age-related diseases have not yet been conclusively demonstrated. Most human trials have been short-term and have focused on safety and biomarker changes rather than hard clinical endpoints. As such, while the theoretical rationale and preclinical data are promising, human evidence remains limited, justifying a moderate evidence rating.
