Polyphenols are a diverse class of naturally occurring compounds found abundantly in plant-based foods such as fruits, vegetables, tea, coffee, and wine. Their use for aging prevention is primarily justified by scientific evidence, though this evidence is mostly preclinical or observational in nature. Polyphenols possess strong antioxidant and anti-inflammatory properties, which are believed to counteract several cellular processes associated with aging, such as oxidative stress and chronic low-grade inflammation ("inflammaging"). Several studies have shown that polyphenols can modulate signaling pathways related to longevity, such as the sirtuin and AMPK pathways, and may improve mitochondrial function and DNA repair mechanisms.
Epidemiological data suggest that diets rich in polyphenol-containing foods are associated with a lower risk of age-related diseases, including cardiovascular disease, neurodegenerative disorders, and certain cancers. Some randomized controlled trials have found that specific polyphenols (such as resveratrol, EGCG from green tea, or flavonoids) may improve biomarkers of aging or reduce risk factors for age-related diseases, though the effects are generally modest and often not directly linked to prolonged lifespan in humans. Most of the compelling evidence comes from animal models or cell-based studies, with limited direct clinical data on aging prevention in humans. Therefore, while there is a plausible scientific rationale and growing body of supportive data, more robust clinical trials are needed to establish efficacy for aging prevention.
