Nicotinamide Mononucleotide (NMN) is promoted as a supplement to support healthy aging, largely based on its role as a precursor to nicotinamide adenine dinucleotide (NAD+), a vital molecule in cellular metabolism and energy production. Animal studies, particularly in mice, have shown that NMN supplementation can increase NAD+ levels, which decline with age, and this has been associated with improved mitochondrial function, enhanced physical activity, and some markers of metabolic health. Key research by Dr. David Sinclair and others has highlighted potential benefits in rodent models, suggesting delayed aging processes and extended lifespan in certain contexts. However, human clinical evidence remains limited and preliminary. A handful of small, short-term human studies have demonstrated that NMN supplementation can increase NAD+ levels and may have modest effects on insulin sensitivity, fatigue, or muscle performance, but robust data showing a clear anti-aging effect in humans is lacking. Major regulatory bodies do not currently recognize NMN as a proven therapy for aging or age-related diseases. In summary, while there is a sound biological rationale and promising preclinical data, clinical evidence in humans is still emerging and the benefits for aging prevention are not yet well-established.
