AMP-activated protein kinase (AMPK) is an energy-sensing enzyme that plays a central role in cellular metabolism, autophagy, and mitochondrial health. Its activation has garnered scientific interest in the context of aging prevention due to its involvement in pathways that mitigate age-related cellular dysfunction. Research in animal models and cell cultures has demonstrated that AMPK activation can extend lifespan and healthspan, largely by promoting cellular processes associated with longevity such as autophagy (cellular cleanup), improved insulin sensitivity, and reduced inflammation.

AMPK activators, such as metformin and resveratrol, have shown promising results in preclinical models, including reduced markers of aging and improvement in age-related diseases. In humans, indirect evidence from epidemiological studies suggests that drugs activating AMPK may be associated with healthier aging, but direct, large-scale clinical trials in non-diseased, aging populations are limited.

While the mechanistic rationale for targeting AMPK in aging is robust, and animal studies are supportive, the translation to human aging prevention is not yet fully established. Thus, the scientific support is moderate (3/5): there is credible mechanistic and preclinical evidence, but human efficacy data are still emerging. Continued research and clinical trials are necessary to confirm its role and effectiveness as an anti-aging intervention in humans.
