Alpha-Ketoglutarate (AKG) is a key intermediate in the Krebs (citric acid) cycle, playing a vital role in cellular energy production and amino acid metabolism. Interest in AKG for aging prevention stems from animal and cellular studies suggesting that it may influence lifespan and age-related processes. A notable 2020 study in the journal Cell Metabolism demonstrated that calcium AKG supplementation extended median lifespan and healthspan in mice, apparently by reducing inflammation and age-related signaling pathways (link). Additional research has indicated that AKG levels decline with age in humans and that supplementation may help offset age-associated physiological decline.

However, robust clinical evidence in humans is lacking. Most supportive data come from preclinical animal studies or in vitro research. There are only preliminary human trials, often with small sample sizes or focused on other endpoints (e.g., muscle metabolism, not direct aging markers).
