Alpha-ketoglutarate (AKG) has gained attention in recent years as a potential anti-aging supplement, primarily based on preclinical research. AKG is a crucial intermediate in the Krebs cycle and plays a role in cellular energy metabolism, amino acid synthesis, and epigenetic regulation. Several animal studies have shown that supplementation with AKG can extend lifespan and healthspan. For example, a 2020 study published in Cell Metabolism demonstrated that calcium alpha-ketoglutarate supplementation extended median lifespan and delayed frailty in aging mice. The proposed mechanisms include reduction of systemic inflammation, improved metabolic health, and modulation of gene expression via epigenetic pathways (such as influencing DNA and histone demethylases).

However, direct clinical evidence in humans is very limited. While AKG is generally recognized as safe, and its physiological roles are well understood, there are no large randomized controlled trials confirming anti-aging effects in people. A few small human studies have explored its effects on muscle metabolism and wound healing, but these do not directly address aging or longevity. As such, while there is promising scientific rationale and preclinical data, the current level of evidence for AKG as an anti-aging intervention in humans remains preliminary.
