Vitamin B5, also known as pantothenic acid, plays a key role in the synthesis of coenzyme A, which is essential for energy metabolism and the production of steroid hormones, including those produced in the adrenal cortex (such as cortisol). Animal studies have shown that pantothenic acid deficiency can lead to impaired adrenal function and reduced synthesis of adrenal hormones. Some older research from the mid-20th century observed that rats deprived of pantothenic acid exhibited adrenal atrophy and reduced ability to respond to stress, which suggested a link between B5 and adrenal health. However, direct high-quality clinical evidence in humans is limited. Supplementation of pantothenic acid is generally only required in cases of deficiency, which is rare, as the vitamin is widely available in foods. While the theoretical and biochemical basis for supporting adrenal cortex function is strong, there is a lack of robust human trials directly demonstrating benefit from supplementation for adrenal health or stress resilience. Thus, the use of vitamin B5 for adrenal support is scientifically plausible and partly based on animal studies, but the evidence in humans is weak, justifying a moderate-low rating.
