Vitamin B complex, particularly B5 (pantothenic acid), B6 (pyridoxine), and B12 (cobalamin), is commonly used to support the adrenal cortex, which is responsible for producing stress-related hormones such as cortisol. The scientific rationale is based on the role of these vitamins as essential co-factors in the biosynthesis of adrenal hormones and in cellular energy metabolism. Research in animals and in vitro models shows that deficiency of B vitamins can impair adrenal function and hormone production. Some small human studies and clinical observations also suggest that B vitamins may help normalize adrenal function in cases of deficiency or chronic stress. However, high-quality, large-scale clinical trials directly demonstrating a benefit of B vitamin supplementation for adrenal support in healthy humans are lacking. Most evidence is indirect or comes from studies of people with deficiencies or high stress loads. Thus, while there is a plausible biochemical basis and some preliminary evidence, the strength of evidence for B vitamins as adrenal cortex supporters in the general population is moderate at best, and supplementation is most clearly justified in cases of deficiency, poor diet, or increased physiological demand.
