Whole adrenal glandular supplements have a traditional basis for their use in supporting or treating Addison's Disease, rather than a foundation in robust scientific evidence. Historically, before the discovery and synthesis of corticosteroids, desiccated and raw adrenal gland extracts from animals were administered to patients with Addison’s Disease (primary adrenal insufficiency) in an attempt to replace the missing adrenal hormones. Some early 20th-century case reports and medical texts describe mild symptomatic improvement in patients given adrenal tissue extracts; however, these preparations were highly variable in hormonal content and often ineffective in delivering adequate hormone replacement. Once cortisone and hydrocortisone were isolated and could be synthesized reliably, they rapidly replaced glandular extracts due to their proven efficacy and safety profile.
Modern scientific studies do not support the use of whole adrenal glandular supplements for Addison’s Disease. There is little to no clinical evidence that commercially available glandular products provide significant or consistent amounts of active adrenal hormones, and their use carries potential risks, such as contamination or transmission of disease. Current medical guidelines recommend only pharmaceutical corticosteroid and mineralocorticoid replacement. Thus, while the use of adrenal glandulars has a place in medical history, it is not validated by contemporary science and is considered obsolete in current evidence-based practice.
