Glycosaminoglycans (GAGs), such as chondroitin sulfate and hyaluronic acid, are naturally occurring polysaccharides that play a vital structural role in connective tissues, including cartilage and bone. Scientific studies have primarily focused on their use in joint health, particularly in osteoarthritis, due to their involvement in cartilage matrix structure and function. Clinical trials have found that chondroitin sulfate supplementation can modestly reduce pain and improve joint function in osteoarthritis patients, suggesting it helps maintain cartilage integrity. There is also evidence that GAGs may influence bone remodeling by affecting the activity of osteoblasts and osteoclasts, the cells responsible for bone formation and resorption. However, the direct evidence supporting the use of GAGs specifically for bone health (as opposed to cartilage or joint health) is more limited. Most benefits appear related to their effect on cartilage preservation, which indirectly supports bone by maintaining joint structure. Some animal studies suggest potential benefits for bone density, but robust clinical data in humans are lacking. Overall, while there is scientific rationale and some supporting evidence for GAGs in supporting the skeletal system, particularly via cartilage and joint health, the evidence for direct effects on bone health is moderate.
