Withanolides are a group of naturally occurring steroidal lactones found primarily in the plant Withania somnifera, commonly known as ashwagandha. There is a growing body of scientific evidence suggesting that withanolides can influence the serotonin system in the body. Preclinical studies have demonstrated that extracts containing withanolides can modulate neurotransmitters in the brain, including increasing serotonin levels. For example, several animal studies indicate that ashwagandha extracts may exert antidepressant-like effects, possibly through serotonergic pathways, as evidenced by behavioral tests and measurements of serotonin in brain tissue. Some studies in humans, while limited in size and scope, have shown improvements in mood and reductions in symptoms of anxiety and depression after supplementation with ashwagandha extracts standardized for withanolide content. However, the direct effect of isolated withanolides on serotonin in humans is not as well established as the effects of the whole plant extract. The exact mechanisms remain under investigation, but potential actions include modulation of serotonin receptors and indirect regulation through stress reduction and HPA axis balance.
