Vitamin B6 (pyridoxine) is commonly used to support or treat premenstrual syndrome (PMS), particularly PMS Type P (predominantly psychological symptoms such as mood swings, irritability, anxiety, and depression). Several clinical studies have investigated the efficacy of vitamin B6 for PMS, with mixed but generally positive results. A 1999 meta-analysis published in BMJ reviewed nine randomized controlled trials and concluded that vitamin B6 is more effective than placebo in relieving overall PMS symptoms, especially mood-related complaints. The proposed mechanism is that vitamin B6 serves as a cofactor in the synthesis of neurotransmitters such as serotonin and dopamine, which are involved in mood regulation. Doses used in studies typically range from 50-100 mg/day, which are higher than the recommended dietary allowance but below the threshold for toxicity. However, more recent reviews note that while there is evidence for benefit, the quality of studies varies, and larger, well-designed trials are needed to confirm efficacy and optimal dosing. Side effects are generally mild at recommended doses but neuropathy can occur at excessive intakes.
