Withanolides are a group of naturally occurring steroidal lactones found primarily in plants of the Withania genus, most notably Withania somnifera (Ashwagandha). Ashwagandha has a long tradition in Ayurvedic medicine for its adaptogenic, stress-reducing, and restorative properties. In recent years, scientific studies have investigated Ashwagandha and its withanolides for their effects on the nervous system, including the parasympathetic branch, which governs "rest and digest" functions.

Preclinical research suggests that withanolides exert anxiolytic and anti-stress effects, partly by modulating neurotransmitters such as GABA and by reducing cortisol levels. Human clinical trials have shown that Ashwagandha supplementation can reduce perceived stress and anxiety, improve sleep, and support homeostatic balance, all of which are linked to parasympathetic nervous system activation. For example, randomized controlled trials (RCTs) have reported significant reductions in stress and anxiety markers with Ashwagandha extract supplementation compared to placebo.

However, most studies investigate Ashwagandha as a whole and not isolated withanolides. Direct scientific evidence linking withanolides specifically and exclusively to parasympathetic nervous system support is limited, but the overall body of evidence for Ashwagandha's calming and anti-stress effects suggests a supportive role for withanolides in modulating autonomic balance. The evidence is moderate but not definitive, justifying a mid-range rating.
