8-Prenylnaringenin (8-PN) is a potent phytoestrogen found in hops (Humulus lupulus). Its use for osteoporosis is primarily supported by scientific studies, though most evidence comes from preclinical research and a few small human trials. 8-PN binds to estrogen receptors, particularly ERα, with higher affinity than many other plant-derived phytoestrogens. Estrogen plays a crucial role in bone metabolism, and its deficiency after menopause is a major factor in osteoporosis development.

In animal models, 8-PN has demonstrated the ability to prevent bone loss induced by estrogen deficiency, improving bone mineral density and bone strength. These effects are attributed to its estrogenic activity, which promotes osteoblast activity (bone formation) and inhibits osteoclast-mediated bone resorption (bone breakdown). A few small-scale human studies and observational reports suggest that hop-derived extracts containing 8-PN may have beneficial effects on bone turnover markers in postmenopausal women, though robust clinical evidence is lacking.

While traditional use of hops has focused more on sedative and menopausal symptom relief rather than osteoporosis, the scientific rationale for 8-PN's bone-protective effects is plausible and supported by preliminary data. However, more large-scale and long-term clinical trials are needed to firmly establish its efficacy and safety in osteoporosis management.
