Pantothenic acid (vitamin B5) is essential for the synthesis of coenzyme A (CoA), a critical cofactor in mitochondrial energy metabolism. CoA is required for the Krebs cycle (also known as the citric acid or TCA cycle), which occurs within the mitochondria and is fundamental to cellular energy production via ATP synthesis. Deficiency in pantothenic acid can impair CoA production, potentially disrupting mitochondrial function. However, in healthy individuals with adequate dietary intake, pantothenic acid deficiency is rare. Most scientific studies confirm its indispensability in basic mitochondrial function, rather than demonstrating improved mitochondrial performance or energy levels with supplementation above the recommended dietary allowance.
There is little robust clinical evidence supporting the use of pantothenic acid supplementation to enhance mitochondrial health or function in individuals without a deficiency. The majority of evidence comes from animal studies and biochemical knowledge, not from clinical trials in humans targeting mitochondrial disorders or dysfunction. Thus, while pantothenic acid is biochemically required for mitochondrial energy metabolism, there is only limited scientific support for using it as a supplement specifically to "support the mitochondrial body system" in people who are not deficient.
