Specialized pro-resolving mediators (SPMs) are bioactive lipid compounds derived from omega-3 fatty acids. They include resolvins, protectins, and maresins, and are involved in actively resolving inflammation. SPMs have attracted scientific interest for their potential role in supporting joint health, particularly in conditions characterized by chronic inflammation, such as osteoarthritis and rheumatoid arthritis.

Preclinical studies (in vitro and animal models) have demonstrated that SPMs can reduce inflammatory cell infiltration, decrease production of pro-inflammatory cytokines, and promote the clearance of inflammatory debris within joint tissues. These actions are distinct from traditional anti-inflammatory drugs, as SPMs do not merely suppress inflammation but actively promote its resolution and tissue repair.

Human clinical research is still at an early stage. Some small pilot studies and case reports suggest potential benefits of SPM supplementation in reducing joint discomfort and improving function, but large, well-designed randomized controlled trials are lacking as of 2024. Nevertheless, the mechanistic rationale and early clinical signals provide a moderate level of scientific support for SPMs in joint health, especially in the context of chronic inflammatory conditions. Further research is needed to confirm efficacy, optimal dosing, and safety.
