Vitamin B6 (pyridoxine) has traditionally been used to support mood and emotional well-being, including symptoms such as irritability. This use originates from observational associations between B6 deficiency and a range of neuropsychiatric symptoms, including irritability, depression, and confusion. Historically, clinicians noted that correcting B6 deficiency in certain populations (such as those with malnutrition or specific metabolic disorders) could alleviate irritability and related symptoms.
However, while there are a few small studies suggesting that B6 supplementation might have a modest effect on premenstrual syndrome (PMS)-related mood symptoms (including irritability), the overall scientific evidence is limited, inconsistent, and not robust enough to justify widespread use for irritability in the general population. Most clinical guidelines do not recommend B6 supplementation for irritability except in cases of confirmed deficiency.
